CBD and The Endocannabinoid System

Israeli chemist Dr. Raphael Mechoulam discovered some active materials in the cannabis plant. CBD was discovered in 1963 and THC in 1964.

The journey of research to determine how these molecules work led to the discovery in 1990 of a human system, deep within, called the ECS (endocannabinoid system). The ECS works where human message transfer takes place. These messages are transferred at the end of a neuron across a space called the synaptic cleft.  These messages called neurotransmitters are both electrical and chemical.

As you will see, the ECS is affected by the phytocannabinoids (CBD, THC) and greatly affected by molecules we make in our bodies, called endocannabinoids (anandamide and 2-AG).

The CB receptors.

In our body are 2 receptors sites where the phytocannabinoids (CBD,THC) and endocannabinoids (anandamide, 2-AG).

 

These receptor sites are aptly named CB1 and CB2.

 

These molecules and these sites are the main players along with enzymes, such as FAAH.

 

CB1 is primarily expressed in the brain.  It exists on the neuron at the synapse.

 

CB1 is the most abundant G coupled receptor in the brain and has major neuromodulatory functions.

THC fits into the CB1 receptor like a lock and key mechanism. Our natural endocannabinoid anandamide activates this same CB1 receptor.

CBD does not fit into the CB1 neuron receptor the same way as THC. Until very recently many were saying CBD can not attach to the CB1 receptor directly. New science clarifies this.

 

 

The Bliss Molecule

More than two decades after the phytocannabinoids in hemp were discovered the endocannabinoid anandamide was found. Yet, once they isolated the chemical, they faced another challenge. What should it be called? They turned to Sanskrit. Anandamide comes from the Sanskrit word Ananda, which means bliss. So, basically, anandamide means bliss molecule.

Since the initial discovery of the CB1 receptor site by Allyn Howlett and William Devane in 1988, it has been an “accepted” fact that CBD, unlike THC, has little binding affinity for the CB1 receptor. Unfortunately, this assumption was not based on science. New data emerging from the international cannabinoid research community indicates that CBD interacts directly with the CB1 receptor site in ways that are therapeutically relevant. CBD parks at a different docking site on the CB1-R that is functionally distinct from THC’s orthosteric binding site. CBD attaches to what’s known as an “allosteric” binding site on CB1-R. When CBD docks at the receptor, it does not initiate a signaling cascade. It does, however, influence how the receptor responds to stimulation by THC and the endogenous cannabinoids. Allosteric modulation of CB1-R changes the conformation (shape) of the receptor, and this can have a dramatic impact on the efficiency of cell signaling.

CBD can chaperon this neurotransmitter within the ECS.

 

Thus CBD enhances neuronal message transmission without the intoxicating effect of THC. You do not have the intoxicating side effects created by THC which are short term memory loss and motor function impairment.
